A multi-tier architecture typically involves the separation of presentation, business logic, and data management layers. Using a read replica with RDS primarily addresses scalability and availability within the data layer by allowing read traffic to be distributed across multiple copies of the database, but it does not by itself constitute a multi-tier architecture which would include separate logic and presentation layers as well.
